I believe the thing you are talking about is a dlc item for those who bought the limited edition version of the game. if you open it you get one of the best swords in the game the wolfbane. its a quest basically, hope that helps

There is a "Relationship Quest -- Courier" to "Take the package to Anne." in Silverpines. The glowing trail leads to a door which kinda glows green, but does not open. Nor does it indicate anything about a key (ie: This door is locked. or This door will not open without a key.)
